چکیده

Urinary 8-oxo-7,8-dihydro-2‟-deoxyguanosine (8-oxodGuo) is widely used as a marker of oxidative stress. Herein we report the comparison of two, distinct chromatographic assays, with ELISA. Chromatographic assays displayed good agreement (r = 0.89, p < 0.0001), whereas there was markedly worse, albeit still significant, agreement with ELISA (HPLC-GC/MS: r=0.43, HPLC-EC: r=0.56; p<0.0001). Mean values differed significantly between chromatographic assays and ELISA (HPLC-GC/MS: 3.86, HPLC-EC: 4.20, ELISA: 18.70 ng/mg creatinine, p<0.0001). Whilst it is reassuring to note good agreement between chromatographic assay, this study reveals significant short-comings in the ELISA, which bring into question its continued use in its present form.

Toward consensus in the analysis of urinary 8-oxo-7,8-dihydro-2'-deoxyguanosine as a noninvasive biomarker of oxidative stress.

Of the DNA-derived biomarkers of oxidative stress, urinary 8-oxo-7,8-dihydro-2'-deoxyguanosine (8-oxodG) is the most frequently measured. However, there is significant discrepancy between chromatographic and immunoassay approaches, and intratechnique agreement among all available chromatography-based assays and ELISAs is yet to be established. This is a significant obstacle to their use in larg...

Analysis of urinary 8-oxopurines 1 Analysis of urinary8-oxo-7,8-dihydro-purine-2’-deoxyribonucleosides by LC-MS/MS and improved ELISA

Non-invasive monitoring of oxidative stress is highly desirable. Urinary 7,8-dihydro8-oxo-2’-deoxyguanosine (8-oxodG) is a biologically relevant and convenient analytical target. However, immunoassays can over-estimate levels of urinary 8oxodG. Measurement of more than one DNA oxidation product in urine would be advantageous in terms of mechanistic information. Urines samples were analysed for ...

The effect of the 2-amino group of 7,8-dihydro-8-oxo-2′-deoxyguanosine on translesion synthesis and duplex stability

Replication of DNA containing 7,8-dihydro-8-oxo-2'-deoxyguanosine (OxodG) gives rise to G --> T transversions. The syn-isomer of the lesion directs misincorporation of 2'-deoxyadenosine (dA) opposite it. We investigated the role of the 2-amino substituent on duplex thermal stability and in replication using 7,8-dihydro-8-oxo-2'-deoxyinosine (OxodI). Oligonucleotides containing OxodI at defined ...

Urinary 8-oxo-7,8-dihydro-2'-deoxyguanosine values measured by an ELISA correlated well with measurements by high-performance liquid chromatography with electrochemical detection.

Measurement of urinary 8-oxo-7,8-dihydro-2'-deoxyguanosine (8-oxo-dG) has recently become more popular as a means of assessing oxidative stress in the human body. Although using high-performance liquid chromatography with electrochemical detection (HPLC-ECD) is a reliable method of measuring 8-oxo-dG, easier and simpler alternative methods may be preferred, if they are quantitatively accurate. ...

Assessing Free-Radical-Mediated DNA Damage during Cardiac Surgery: 8-Oxo-7,8-dihydro-2′-deoxyguanosine as a Putative Biomarker

Coronary artery bypass grafting (CABG), one of the most common cardiac surgical procedures, is characterized by a burst of oxidative stress. 8-Oxo-7,8-dihydro-2'-deoxyguanosine (8-oxodG), produced following DNA repairing, is used as an indicator of oxidative DNA damage in humans. The effect of CABG on oxidative-induced DNA damage, evaluated through the measurement of urinary 8-oxodG by a develo...
